From Santiago Airport you have 2 low cost alternatives (besides just renting a car or taking a cab for US$110)
- If you're arriving in LAN, they'll arrange a reservation for their transfer (US$20 return pp) which leaves from the airpors and leaves you in Viña del Mar.
- If you're not, or would rather take the experience, take the Centropuerto or Tur Bus to the Pajaritos Metro Station (US$3) and from there take a bus to either Viña or Valparaiso (about US$5 depending on the season)
The hotel I'd recommend is the Hotel del Mar, IME the best hotel in Viña del Mar, attached to the Casino and right across from the sea. Other hotels I'd recommend are the Radisson Acqua in Reñaca or the Sheraton Miramar, very close to the Hotel del Mar.
In Valparaiso, I've gotten really good appreciations about the Zero Hotel, although this is a boutique style hotel, and maybe not your type. There's going to be an opening from a Diego de Almagro hotel soon in Valparaiso.
At the hotels they'll offer you a vast range of tours and activities to the vineyards, national parks, etc...
I'd say not to miss Maitencillo beach, as I believe this is one of the best beaches in the surroundings.

You can take a Private vehicle and on the way visit some vineyards.
If you like nice hotels I would reccomend Zero, Casa Higueras, Somerscales or Acontraluz. Explore the port for 2 nights and the head to Viña lodging in Sheraton or del Mar and then the last night in Con Con lodging at Radisson Acua.
Let me know if you want me to tailor made a private tour for you.

A must see is La Campana national park, there are plenty of places to explore and you can take a dip in a mountain fed spring there. Also check out Horcon or Laguna Verde as described in my profile. We specialize in these types of tours so let me know if you see anything that interests you. I will be updating my services in the next few weeks so stay tuned.
As for transpotation I think it would be best to follow the advice of Matias and if you opt to take local buses just make sure your spanish is good.
One hotel which was not mentioned is hotel San Martin in Viña. My parents stayed there when they visited, it was really nice and had a great breakfast.
I hope you guys have fun on your trip chau chau
