If you are coming by yourself, then your best choice would be to get a bus from Tres Cruces to Punta del Este. The companies that will take you there are named COT and COPSA. Make sure that you buy a DIRECT bus ticket, because otherwise the trip can take up to one hour extra.
I would recommend that once you're in Punta Del Este you rent a car for the day or hire a tour, because it is a pretty vast place with many interesting places to visit; the only problem is that they are a bit apart from each other.
My recommendations for MUST SEE sites are: Casapueblo, Gorlero Avenue, seaside walks, José Ignacio and the Ralli museum.

I totally agree with answer 1 given by Natalia. The cost of the bus tickets COT or COPSA is approximately 160 uruguayan pesos one way (US$ 8.5).
I live in Punta del Este all year round and the city and its surroundings are absolutely beautiful. In March, when the high season has passed and the weather is cooler, you can still enjoy the beaches and a swim in the sea.
The offers are very varied, so where you go depends on what you prefer if you are coming for only one day: Nature trips (Arboretum Lussich, Lapataia, beaches, Museo del Mar), city tour or rent a car and just drive around the towns sightseeing (Punta Ballena, Punta del Este, La Barra, Manantiales, José Ignacio) or just arrive and walk around Punta del Este and enjoy the beaches (Brava and Mansa) and the port. Hello,you can take a bus in Tres Cruces,and take a coty tour or something in the bus station in Punta del Este.I,m going to punta del este every week.If you want,contact me and we can go together.

Natalia made the best response in my opinion,
Make sure to ask for the "DIRECT bus" (Directo Punta del Este)
You do not have to book in advance, just drop by to "TRES CRUCES" bus terminal and buses leave every 15/20 minutes...
The best thing to do is walk along the beach near the port (Peninsula). It is beautiful.
Try to go to "la barra", you have to cross a "special" bridge, try to acelerate while crossing it! it is fun!
Once there rent a car o take a excursion.

What about going to Rocha? It is very nice as well, but different...more natural.

hi there!!, if you wanna go by bus you can take a direct bus from the bus station called "Tres Cruces" there you will find two bus company that will take you to Punta del Este. the companies names are COT and COPSA both are good and quiet cheap .

Try buquebus, from the web you can reserve the ticket to travel to punta del este and from punta back to montevideo

How are you? I would recommend you to take a Copsa bus on the bus terminal called "Tres Cruces". They have brand new comfortable buses even with wi fi internet. March is a great month to go to Punta, since weather is really nice and the city it´s not too crowded. The ticket is about $160 pesos.

Hi Mariela,
The recommendation to come by bus of my colleagues is one option, another one is to come by private transfer (maybe if you come for the day you want to optimize your time), in that case you can book a car for USD 170, or rent one in Montevideo and come to Punta driving, this is cheaper, you will pay USD 100 for the entire day, and the route is very nice and safe.
Once in Punta we can take you in a tour to visit the best places.

We have dayly services at 8h departure with other people in bus and the price is 40 U$ contact me and you must make reservation with me and wait the bus in the lobby of any hotel in the downtownt in montevideo.
