Madrid is not my specialty, but it's a great city to visit!
So I can give you a few ideas:
MUSEUMS: BIG thing in Madrid! Don't miss the Prado, the Reina Sofia and the Thyssen. Oups... Just this three could easyly fill up your whole 2.5 days!
Parks: El Retiro. You wouldn't go to NYC and miss Central Park, right?
Landmarks: Puerta de Alcalá, Cibeles fountain, Plaza Mayor, Puerta del Sol, Gran Via, Calle de Alcalá.
Buildings: Palacio Real, Catedral de la Almudena, Real Madrid stadium, Teatro Real.
I'm sure I'm missing many fun places, but I haven't been there for a while. It's too bad you won't have time for the many great daytrips around Madrid. The Monasterio del Escorial is a MUST!
My last tip... If you wish to use your time in the most efficient way, hire a private tourguide even if it's just for a half day. He/she will be able to create the itinerary that makes most sense for the time available, skip lines to enter museums and sites and will give you plenty of good ideas and advice to organize the rest of your time in Madrid. No comparison with a big step-in group and light years difference with the hop-in buses! Nothing beats personal attention!

For me the most interesting and beautiful places in Madrid are the Museum (El Prado, Reina Sofia, Thysen, Caixa Forum, etc). During nights there are lots of good bars, restaurant, pubs, etc. I strongly recommend you the visit of Museum.

I think I nhve answerd this question already. Anyway you may also spend a day in Toledo or Segovia both towns are worth a visit and can be reached by the AVE high speed train in just 35 minutes. From the train sation you have to take a bus to the city center. AVE trains leave from the nice Atocha train station. Another visit may be the Royal palace, botanical garden and Retiro park in Madrid.
