Singapore is relatively small hence most of the local sights are spread out throughout the island.
I would feel that either Bugis or Orchard would be the most accessible.
For Bugis, you can explore cultural areas like
Little India, Geylang Serai, Chinatown(four stations away).
Within Bugis itself, there are many shopping sites and occasional exhibitions and events at the Suntec city which is a stone's throw away.Sentosa island (nearest MRT is Harbour Front Station) is a must visit. For 4 -Day visit, I would recommend the following itinerary

Day 1
1) Explore the surrounding areas near your hotel and try the local food. Sometimes the places that are closest to our hotel get missed out until the day we have to leave home.
2) Day 2
Take a train to Harbour front Station, have breakfast at the Seah Im Food centre just right next to the Station or at Vivo City-Food Republic.

Board the bus to Sentosa island and spend the rest of the day there. You can explore places like the Underwater world,Wax museum, Butterfly park,have a pinic lunch at the Siloso beach( food can be bought at Giant Super market at Vivo city). Enjoy the Merlion & musical fountain before heading back to the main island. Dinner can be had in Vivocity or if you prefer to try lcao food, you can take the MRT to City Hall station and have your meal at the Esplanade-Makan Sutra Food Court. Take a stroll along the Esplanade or take the Singapore River Boat Cruise.

Day 3
Explore the Western park of Singapore and go to the Singapore Bird Park, Science centre, Snow city and try Prawn fishing near the Singapore Bird Park. Take a train and visit the Night Safari(but my take is, it's not really worth it). The Singapore Zoological gardens are much better which you can visit in Day 4.
Day 4.
Singapore Zoological gardens or last minute shopping at Bugis-Little India area and head to the airport.

Hotels near Bugis
No frills: Beach Hotel(near Suntec city and parked near where most of the eateries are along Bugis).Friendly staff.
Hotel81-Bencoolen, rather small but a TV, bed and the pillows are good.
The new iBis hotel but I have not stayed there so cannot recommend about the conditions.
Orchard, I think there is a 4* hotel called the Grand Central Hotel which is behind the Concord hotel, easily accessible to Plaza Singapura, Orchard Road shopping malls etc.
If you are looking for a local tour, you can google Geeraldene Lowe who does great walking tours with interesting snippets of Singapore History.

I would suggest the budget Hotel 81. Its reasonably priced & you get clean rooms equipped with A/C,TV,shower with heater,hairdryer,kettle,beverages. Prices range from S$60 - S$159(depending on the location). The Hotel 81-Chinatown is easily accessible to other parts of Singapore by mrt. D Chinatown mrt is just across the road. Smith Street is nearby,it has a wide variety of Singaporean "hawker" fare. Vendors there are specially selected for their culinary skills. Prices are displayed & reasonably priced. Peoples' Park Centre,Peoples' Park Complex & Chinatown Point with lots of shops are across the road too. Pagoda & Mosque Streets are also good for shopping.
Getting to Sentosa is accessible from Chinatown mrt.
The cheaper hotels are found along Joo Chiat Road or along the Lorongs of Geylang. Think it'll be good if you can go to the website : regarding the location & prices.
In fact transport to every part of the island is very convenient: either by buses,taxis or the mrt.
Where activities are concerned,it all depends on the individual.

For hotels is suggest you contact the travel agent. They normally have a much better rate as compared to 'walk in rate'. Try quality hotel at ballestier, its cheap and reasonable hotel. As for activity, can visit Singapore Day Zoo, night safari, walk around china town for food and shopping and also Sentosa. Make sure they watch the "song by the sea"
