Go to Sonoma and Napa- up the street over the Golden Gate Bridge. My all time favorite is V. Sattui Winery. You can only buy their wine from the Vineyard or online. They also have an AMAZING deli with food that is out of this world. It is busy there but don't let this fool you, the lines move fast and it is well worth the things they have to offer.
What about transportation... are there tour buses, guides... we plan to leave in the morning and return in the evening
Sonoma will be closer; both have great wines, depending upon your tastes. On the Napa side, Domaine Chandon has great sparkling, for big reds try Signorello, Robert Sinskey, Steltzner. Great tasting rooms in Yountville are Girard, Hope & Grace, Hill Family. If you have specifics let me know.
Well, I would say go to Sonoma as it's less commercial but.... V. Sattui is VERY popular but VERY crowded and, if you're a wine afficianado I would say there are better places to go. Sonoma Square is very charming and a couple of wineries not too, too far away that are gorgeous and smaller are Cline...Hwy. 121 on the way to Sonoma Sq.(Hwy. 12), Domaine Carneros(Napa Rd.), which has a lovely sparkling and terrace overlooking the vineyards...nice pinot too. Also in Sonoma...Valley of the Moon, Kunde, Chateau St. Jean(gorgeous gardens & good chardonay). A nice small winery near Domaine Carneros...well, two...are Acacia(needs a res, I think) and Bouchaine, which does not. You can Google these wineries for good directions.
In Sonoma, have lunch at the Swiss Hotel or The Girl and the Fig or any one of a number of cute other places.
Be aware that Saturday is THE most crowded day up there so, if you can make it on another day, I would do that. If not, stay away from the huge wineries....no fun fighting the crowds.

Cheers!
